About this role
Your responsibilities: Characterization of biopharmaceutical products, including monoclonal antibodies, multi-specifics, antibody-drug conjugates (ADCs) and antibody-oligo conjugates (AOCs), their post-translational modifications and degradation products using state-of-the-art instrumentation. Optimization of sample preparation, liquid chromatography (LC), ion mobility separation (IMS), and mass spectrometry (MS) acquisition methods for the characterization and monitoring of product quality attributes. Writing of method development and robustness reports. Writing/review of MS sections in characterization reports for the structure elucidation of drug substance, characterization of reference material, degradation products and impurities. Communication of results at internal meetings and international conferences. Promote environmental sustainability and GreenLab initiatives in daily work. Maintenance and qualification of lab equipment. LC-MS instrument troubleshooting and technical liaison with instrument vendors. Documentation of experiments and structured data in electronic systems. Minimum Requirements: MSc in Biotechnology, Biochemistry, analytical chemistry or related field. Strong communication skills in English; proficiency in German or French would be an asset. Strong hands-on experience in liquid chromatography (HPLC and UHPLC e.g. Thermo Vanquish and Waters Acquity systems). Hands-on experience in performing MS analyses of biopharmaceutical products at intact, subunit and peptide level using e.g. reversed-phase chromatography with MS (RP-MS), cation-exchange chromatography with MS (CEX-MS) and Multi-Attribute Method (MAM). Hands-on experience in protein sample preparation and previous use of automated liquid handler. Strong theoretical knowledge of protein biochemistry and protein post-translational modifications. Theoretical knowledge and previous experience working with mass spectrometers (e.g. Orbitrap, Q-ToF mass analyzers), Data Dependent Acquisition (DDA). Experience with ion mobility separation (IMS) on a Waters cyclic IMS mass spectrometer would be an asset. Experience with collision-induced dissociation (CID), Higher-energy collisional dissociation (HCD), Electron Capture Dissociation (ECD) and/or Electron Transfer Dissociation (ETD) fragmentation techniques would be an asset. Experience with software solutions for intact mass deconvolution, protein identification and semi-quantification of product quality attributes (e.g. Genedata Expressionist Refiner MS). Experience with MS analysis of oligonucleotides would be an asset. Ability to interpret and present scientific information critically and objectively. Proactive and curious mindset to new technologies/methodologies and environmental sustainability, strong organizational skills.
